There were no breaks for the bathroom or water, bring water or buy when you enter as there is a bathroom when you get in, recommend using it. It is busy and everyone is headed the same way through whichever areas they have deemed to have open at that time. The best way to describe it is imagine when you are leaving a venue like a concert or sports event and everyone is trying to leave and squished through the exits, it is that for 2 hours as you go through the museum. There is no AC either so it is hot! The only place with ac is the lobby when you enter and the Sistine chapel. People were very disrespectful and talked the whole time in the chapel and taking pictures, both of which are not allowed, staff were constantly telling people to stop doing both. Even some of the tour guides were talking. Overall very over rated. Saint Peters basilica was much more impressive and had AC! We were going to go up top but we're too hungry and dehydrated to attempt. I'd say just get skip the line tickets and bring water and snacks and take your time to really get to see stuff, rent audio tour if you want history info, but forget a rushed tour.

The Sistine Chapel is almost at the end of the Vatican Museums route. If you get tired, be sure to take the detour, a shorter route avoiding many intermediate museum rooms that are just as suffocatingly full of people and without offering the possibility to sit down and rest. Don't worry if you need to relieve yourself, there are toilets scattered around.
On the occasion I would like to note that all the places we visited in Rome had a lot of tourism. The whole city is flooded with tourists even though it was low tourist season.
Don't make the mistake of traveling to Rome without pre-booking tickets for the places you want to visit, otherwise you won't be able to get in. The queues and waiting are too long. Keep in mind that for almost all attractions you need to pay a ticket and almost all places are checked similar to that of an airport.
In my opinion, the Roma pass is not worth it as it is not like in other cities that allows you almost unlimited access to museums, etc.
